چکیده

A computer model of the fungal colony that grew consuming limited amount of nutrient with three control parameters, growth rate, and nutrient level and diffusion, was constructed to make a basic morphology diagram of mycelial colony. To apply the diagram with those parameters to the mycelial growth of Aspergillus nidulans wild and mutant strains, the relation between the colony expansion and the nutrient level was measured. Differences in the colony patterns of these strains at low nutrient level were attributed to the change in the above relation that caused a shift of the corresponding patterns in the diagram.

Macroautophagy-Mediated Degradation of Whole Nuclei in the Filamentous Fungus Aspergillus oryzae

Filamentous fungi consist of continuum of multinucleate cells called hyphae, and proliferate by means of hyphal tip growth. Accordingly, research interest has been focusing on hyphal tip cells, but little is known about basal cells in colony interior that do not directly contribute to proliferation. Here, we show that autophagy mediates degradation of basal cell components in the filamentous fu...

Quantifying Two-Dimensional Filamentous and Invasive Growth Spatial Patterns in Yeast Colonies

The top-view, two-dimensional spatial patterning of non-uniform growth in a Saccharomyces cerevisiae yeast colony is considered. Experimental images are processed to obtain data sets that provide spatial information on the cell-area that is occupied by the colony. A method is developed that allows for the analysis of the spatial distribution with three metrics. The growth of the colony is quant...

Inoculum standardization for antifungal susceptibility testing of filamentous fungi pathogenic for humans.

Two methods of inoculum preparation for filamentous fungi were compared: counting with a hematocytometer and spectrophotometric adjustment. One hundred eighty-two filamentous fungi pathogenic for humans were used. Colony counts were done for all inoculum preparations. The agreement between the hematocytometer counts and the colony counts (CFU per milliliter) was 97.2%. The reproducibility betwe...

Modelling fungal colonies and communities: challenges and opportunities

This contribution, based on a Special Interest Group session held during IMC9, focuses on physiological based models of filamentous fungal colony growth and interactions. Fungi are known to be an important component of ecosystems, in terms of colony dynamics and interactions within and between trophic levels. We outline some of the essential components necessary to develop a fungal ecology: a m...
